How it works
Cortagen is a synthetic tetrapeptide with the sequence Ala-Glu-Asp-Pro, abbreviated AEDP, listed in PubChem as H-Ala-Glu-Asp-Pro-OH with molecular formula C17H26N4O9.[1] It belongs to a category of short peptides sometimes called bioregulators, a term used mainly in Russian and Eastern European literature for peptides studied for effects on gene expression and tissue-specific biology.
A 2004 paper describing its development states Cortagen was created through directed synthesis based on amino acid analysis of Cortexin, a natural, multi-component peptide preparation from brain cortex tissue.[2] As a single, defined sequence, Cortagen is chemically simpler than Cortexin, but that does not make its clinical effects better established.
Benefits & side effects
Cortagen's evidence base is overwhelmingly preclinical. In a rat sciatic-nerve-injury model, intramuscular Cortagen increased nerve-fiber growth rate and conduction velocity during regeneration.[4] Mouse studies of locomotor activity and anxiety-related behavior found effects that varied by dose and, at some regimens, appeared anxiogenic rather than calming.[7]
- Rat and mouse brain-ischemia studies examined related neurospecific peptide preparations for antioxidant and neuroprotective activity[5][6]
- An in vitro study found Cortagen induced interleukin-2 gene expression less strongly than two related bioregulator peptides[13]
- A 2015 study of lymphocytes from adults over 80 examined effects on age-related chromatin structure, offering early human-cell context, not a clinical trial[14]
No large, modern human trial of Cortagen was identified. Claims that it improves memory, treats dementia or brain injury, or reverses cognitive decline are unproven extrapolations from animal and cell-based findings.
