How it works
DSIP, or delta sleep-inducing peptide, is a small nonapeptide, built from nine amino acids, first reported in a 1977 paper describing a substance isolated from rabbit cerebral venous blood that appeared to trigger delta-wave EEG activity associated with deep sleep.[2] PubChem lists it as a defined chemical entity, but reviewers note that "DSIP-like immunoreactivity" detected in later assays may sometimes reflect related fragments rather than the intact peptide, complicating the broader literature.[3]
Despite its name, DSIP is not an established sleep medication. It remains primarily a decades-old research subject rather than a compound with a modern, well-characterized clinical profile.
Benefits & side effects
The original evidence for DSIP comes from rabbit and rat studies examining EEG delta-wave activity and sleep-stage changes.[2] Human research followed, but consists mainly of small, older pilot and double-blind studies measuring sleep latency, sleep efficiency, and subjective sleep quality in people with poor sleep or chronic insomnia; a 1986 review described the findings as inconsistent and hard to generalize.[3]
A search of ClinicalTrials.gov shows no modern, large-scale DSIP trial program, which limits confidence in current claims.[7] DSIP has also been explored in older, exploratory literature on cortisol and growth hormone signaling, pain, and opioid withdrawal, but these remain mechanistic or anecdotal.[3] Popular online claims that DSIP promotes lucid dreaming are not supported by any identified clinical evidence.
