How it works

PT-141 is the widely used research and marketing name for bremelanotide, a synthetic peptide that acts as a melanocortin receptor agonist. In the United States it is the active ingredient in Vyleesi, a prescription drug the FDA approved in 2019 for a narrow indication: acquired, generalized hypoactive sexual desire disorder (HSDD) in premenopausal women.[1] That approval is specific to Vyleesi's formulation, dose, and labeled population — it does not extend to every product sold online under the "PT-141" name.

This distinction matters because bremelanotide is chemically related to earlier melanocortin research compounds studied for pigmentation and sexual response, and "PT-141" is often used loosely online to describe unregulated peptide vials marketed for broader libido or performance claims that fall outside the approved label.

Benefits & side effects

The core evidence for bremelanotide comes from two randomized, placebo-controlled Phase 3 trials, often referred to as the RECONNECT studies, in premenopausal women diagnosed with acquired, generalized HSDD.[2] Across both trials, bremelanotide produced statistically significant improvements in validated measures of sexual desire and related distress compared with placebo. It did not, however, meaningfully increase the number of satisfying sexual events — a distinction the FDA label itself highlights.

Evidence for use in men, postmenopausal women, or as a general performance enhancer is comparatively thin, resting mainly on earlier mechanistic and animal research on melanocortin agonists rather than dedicated approval-grade human trials in those populations.
